Ok, I put together a new system with a NZXT X62 cooler, NZXT Manta and an Asus Strix Z270I.

The board has a CPU-FAN header but no CPU_OPT (PWM), only other CHA_FAN headers (PWM).

The Manta case has a PWM hub (with an input lead that is supposed to go to the CPU_FAN header - and the X62 has a 'pump tach' 3-wire that is supposed to go to the CPU_FAN header.

There is a lead from the pump head that connects to the fan wire (2 on the rad).

Problem is I don't get any change in the fans on the rad when I select a change from silent to performance.I know something isn't connected right but I can't quite get my head wrapped around it?

In the ASUS software, if there is a picture marked Chassis Fan, click on that to get the next dialog box. In that dialog box, look for a tab box marked Source. Check to see if the Source is set to CPU rather than Motherboard. If its set to CPU then your rad fans should speed up quickly as CPU temp increases.
