And I want it to be enabled for all things (desktop, app, games and so on)
Well, profile should be "tailored" to a specific calibrated monitor.
Anyway, you CAN NOT have enabled the profile for ALL applications. You just "attach" it to your monitor (screen propertios I presume, I don't have Vista) and applications which support it just use the profile, or you can tell them to use the current monitor colour profile (Photoshop, Firefox 3...).
